The Hoffan Drug was a compound created by the Hoffan people in the Pegasus galaxy with the help of the Atlantis Expedition.

The drug was created by the scientists of Hoff over many generations due to the constant Cullings by the Wraith. Working on their predecessors knowledge, newer generations of scientists would use their research in creating a compound that would make their race immune to the Wraith's ability to feed on them. They were able to perfect the compound through the aid of the Atlantis Expedition's Doctor, Carson Beckett. It was tested on a terminally ill Hoffan who was left in a room with a captured Wraith, called Steve, who attempted to feed on him but broke away from the action. After this, Steve's internal organs shut down as he was poisoned by the drug and died. The volunteer also died as a result. Seeing this as a success, the drug was administered to the Hoffan population, however, the Atlantis Expedition realized that the drug had a 50% mortality rate resulting in many who were inoculated dying. The Wraith eventually learned of the Hoffan Drug and exterminated the Hoffan race for the threat it represented, leaving Hoff in ruins. From the ancient vaults where the Hoffan scientists kept their research, Michael Kenmore came and looted the area of data relating to the compound. Using a clone of Carson Beckett, Michael perfected the drug and began to subtly distribute it to various Human worlds in the Pegasus galaxy as part of an opening volley in his personal war against the Wraith. Many Wraith Hives that engaged in cullings found themselves poisoned and dying after feeding on these Humans. In addition to this, the Human worlds began to suffer from a plague of deaths caused by the drugs mortality rate which resulted in 30% of those administered by the drug dying in the process. It was believed that Michael's forces distributed the drug through food or water on various worlds in his hopes of collectively weakening the Wraith by targeting their food sources. In a now alternate future (changed when Teyla was rescued and Michael was "apparently" killed by the Daedalus) Michael increased his use of this drug on human worlds and it weakened the Wraith further and caused more in-fighting than ever. After the defeat of Michael in the Battle of M2S-445, the use of this drug on human populations ended, but possibly hundreds of worlds were affected by the drug. (ATL: "The Last Man")
It is revealed that at least one Wraith Hive Ship had begun exterminating worlds holding survivors of the drug. It came to a world where people who survived were resettled and the Wraith Commander tried to force the people of the planet to give up the survivors for presumed extermination. It is revealed that a Wraith Scientist (now dead) was working on a way to detect survivors of the Hoffan drug, but he died when he believed he succeeded and was proven wrong when he fed on a survivor. Doctor Carson Beckett was kidnapped to finish his work and did complete it, but used the discovery of the fact that he too had the Hoffan drug in him to kill the Wraith Commander and escape. The finished detection system was destroyed by Beckett and McKay, leaving the Wraith again unable to detect survivors. (ATL: "Outsiders")
